Course Contents

Plant tissue culture laboratory and devices used, related chemicals, areas of use of the technique, and current applications are explained. Plant tissue culture techniques are applied using various organs of sample plant species, and the effects of plant growth regulators on plants are demonstrated comparatively in practice.

Weekly Schedule

1) Totipotency, cell division and elongation in plants
2) History of plant tissue culture and current application purposes of the technique
3) Introducing laboratory parts and teaching the use of related devices
4) Plant growth regulators, working concentrations and storage conditions
5) Aseptic techniques and sterilization practices
6) Medium components; macro and micro elements, other organic substances and buffers
7) Bitkiye uygun besiyerinin seçilmesi, hazırlanması ve sterilizasyonu
8) Midterm Exam
9) Selection and optimization of a disinfection method
10) In vitro germination of seeds and obtaining explants
11) Establishment of cultures using apical or lateral meristems, and shoot multiplication
12) Establishment of callus cultures from leaf explants and properties of callus tissue
13) Establishment of cell suspension cultures and calculation of cell density
14) Introduction of synthetic seed production stages
15) Quantitative and qualitative evaluation of culture data
16) Final Exam
